Cytokeratin 14 Antibody (DE-SPK14) Summary

Immunogen
The antibody reacts with the Keratin 14 antigen.
Specificity
DE-SPK14 reacts specifically in immunoblotting with human keratin 14 (50 kD). In tissue sections this mAb provides a positive reaction on basal cells of non-keratinizing stratified epithelia, basal cells and suprabasal cells of the epidermis and gingival, myoepithelial cells and squamous cell carcinomas.

Application Notes
Suitable for frozen sections / cell preparations. Also suitable for paraffin-embedded tissue when using the MICROWAVE method.Working dilution: 1:20, based on the Indirect method, with a normal ABC method the dilution can be 5-10 times higher, but optimal dilution should be tested by serial dilution. Dilute immediately before use with PBS or Tris. Positive control: Glandular

Background

Keratin 14 is a member of the keratin family, the most diverse group of intermediate filaments. It is usually found as a heterotetramer with two molecules of keratin 5, a type II keratin. Together they form the cytoskeleton of epithelial cells. Mutations in the genes for these keratins are associated with epidermolysis bullosa simplex. Keratin 14 has been studied as a prognostic marker in breast cancer. This antibody labels the basal layer of stratifying squamous and non-squamous epithelia. The staining pattern iscytoplasmic. It recognizes basal cell carcinomas and squamous cell carcinomas.
