Background

MCL-1 is a distinct member Bcl-2 family and lacks BH4 domain. It is a ubiquitously expressed protein containing PEST sequences and has 2 isoforms: MCL1L (longer isoforms) functions as an anti-apoptotic factor while MCL1S (shorter isoforms) induces apoptosis. Reports suggest Mcl-1 plays a more profound role among Bcl-2 family members in response to a variety of death stimuli. Mcl-1 is vital for the development and maintenance of T and B lymphocyte, macrophage survival and survival of hematopoietic stem cells. It also functions as a key regulator of neutrophil apoptosis and survival and its expression is regulated by PI3K/Akt and STAT3 pathways. Mcl-1 deficiency results in peri-implantation embryonic lethality. Expression of Mcl-1 is critical for survival of Multiple Myeloma cells and RA synovial fibroblasts suggesting a potential therapeutic target.Myeloid cell leukemia-1 (MCL-1) is a member of the bcl-2 family. The carboxy terminal of Mcl-1 and bcl-2 share significant sequence homology. Expression of Mcl-1 is increased upon exposure of ML-1 cells to various types of DNA damaging agents, along with increases in GADD45 and Bax and a decrease in bcl-2.
