Background
Prokaryotes and lower eukaryotes possess activities that remove the plethora of oxidative DNA base damages produced during normal oxidative metabolism and which have been associated with cancer and aging. The availability of the human genome sequence has precipitated identification of human homologs of Escherichia coli Nei. Human NEIL1 is a DNA glycosylase, that phylogenetically belongs to the Fpg/Nei family, primarily found in the bacterial kingdom and is functionally homologous to bacterial Nei. Thymine glycol, a potentially lethal DNA lesion produced by reactive oxygen species, can be removed by DNA glycosylases. The NEIL1 gene is located on the long arm of chromosome 15 that is frequently deleted in human cancers.
