Background

Par-4, originally identified as an apoptosis inducer in prostrate cancer cells, is a 332A.A WT-1 interacting protein. It consists of a C-terminal leucine zipper domain and two putative conserved NLS domains required for nuclear localization of Par-4 and induction of apoptosis. It functions as a transcriptional repressor and modulates the transcriptional activities of WT1. Expression of Par-4 is down-regulated in various cancer types. Activation of Par-4 in cancer cells results in activation of FasL-Fas-FADD-caspase8 apoptotic pathway and inhibition of NF-kB prosurvival pathway. It utilizes elevated PKA and induces selective apoptosis in cancer cells. Role of Par4 has also been associated with neuronal degradation in Alzheimers disease. Prostate apoptosis response-4 (Par-4) is a 38kD protein. It is a death domain-containing protein whose levels increase in neurons undergoing apoptosis as a result of trophic factor withdrawal or exposure to oxidative and metabolic insults. Par-4 levels are reported to be increased in lumbar spinal cord specimens from patients with amyotrophic lateral sclerosis (ALS)1 further suggesting a role in neuronal degeneration.
