Background

FKHR-7H3 recognizes FKHR (ForKHead in Rhabdomyosarcoma), a 70a protein, which is a member of the forkhead homeotic gene family of transcription factors (FKH/HNF-3). Members of the forkhead family can be classified and grouped by their degree of homology within the 100-120aa region that is necessary and sufficient for DNA binding and binding-site specificity. The FKHR subfamily of forkhead proteins is relatively divergent from the other members of the forkhead transcription factor family. With all members in this subfamily containing a signature insertion of 5 amino acids next to the helix 3 in the middle of the forkhead domain, and lack the well conserved amino terminal KPPY sequence found in most forkhead genes. Recently, it has been shown that members of the FKHR gene family are phosphorylated by Akt and this phosphorylation regulates its cellular localization promoting cell survival.
